Development of PCR-based marker for resistance to Fusarium wilt race 2 in lettuce (Lactuca sativa L.)

چکیده
Resistance to multiple races of Fusarium wilt is a desired trait in lettuce (Lactuca sativa L.) cultivars, as it directly related grower profitability. We focused on its resistance the race 2 pathogen, and analyzed F2 individuals obtained from cross between ‘VI185’ (resistance 2) ‘ShinanoGreen’ (susceptible for genetic mapping locus. According ddRAD-seq analysis genotype prediction each individual by infection assay derived F3 generation, single semi-dominant locus LG1 (qFOL1.2) was found determine resistance, marker designated LG1_v8_117.181Mbp showed complete co-segregation with phenotype. Fine identified PCR-based markers this allowed show that qFOL1.2 were located position 116.468–117.974Mbp LG1. Association phenotypes genotypes 42 cultivars all uniform susceptible while their resistant not always consistent. Nonetheless, only LG1_v8_116.506Mbp identical across 25 cultivars. Thus, we defined broadly available selection 2-resistance Our results provide additional breeding tool 2, can accelerate pyramiding loci program.

Mutants of downy mildew resistance in Lactuca sativa (lettuce).
As part of our investigation of disease resistance in lettuce, we generated mutants that have lost resistance to Bremia lactucae, the casual fungus of downy mildew. Using a rapid and reliable screen, we identified 16 distinct mutants of Latuca sativa that have lost activity of one of four different downy mildew resistance genes (Dm). In all mutants, only a single Dm specificity was affected. Ge...
متن کاملInflorescence Development and the Role of LsFT in Regulating Bolting in Lettuce (Lactuca sativa L.)
Lettuce (Lactuca sativa L.) is one of the most important leafy vegetable that is consumed during its vegetative growth. The transition from vegetative to reproductive growth is induced by high temperature, which has significant economic effect on lettuce production. However, the progression of floral transition and the molecular regulation of bolting are largely unknown. Here we morphologically...
متن کاملToxic effects of copper-based nanoparticles or compounds to lettuce (Lactuca sativa) and alfalfa (Medicago sativa).
The increased production and use of nanoparticles (NPs) has generated concerns about their impact on living organisms. In this study, nCu, bulk Cu, nCuO, bulk CuO, Cu(OH)2 (CuPRO 2005, Kocide 3000), and CuCl2 were exposed for 15 days to 10 days-old hydroponically grown lettuce (Lactuca sativa) and alfalfa (Medicago sativa). Each compound was applied at 0, 5, 10, and 20 mg L(-1). At harvest, we ...
متن کاملDevelopment of RAPD Markers linked to Fusarium Wilt Resistance Gene in Castor Bean (Ricinus communis L)
Wilt, caused by Fusarium oxysporum f. sp. ricini, is the most severe disease of castor bean (Ricinus communis). For the development of marker for wilt-resistant gene, 200 F2 individuals from a cross between the resistant cultivar 48-1 and the susceptible cultivar VP-1 were inoculated with a race of Fusarium oxysporum f. sp. ricini. Bulked segregant analysis was used to search for random amplifi...
متن کاملPhosphomannose isomerase gene for selection in lettuce (Lactuca sativa L.) transformation.
A positive selection system using phosphomannose isomerase was employed for Agrobacterium tumefaciens mediated transformation of lettuce (Lactuca sativa L. var. 'Achát'). It was shown that the mannose-based selection system works very well with the lettuce genotype used, reaching up to 25% transformation efficiency on the medium with 20 g/L mannose and 20 g/L sucrose. The best transformation ef...
